McCallum advanced from 97 not out overnight to a brilliant 181 in the ICC Intercontinental Cup match, Scotland posting a first-innings 452.
Then the attack ripped through the Dutch batting line-up, bowling them out for 138 to claim an emphatic win.
Blain took 3-34, and eight wickets for the match, while Hoffmann took 3-33.
McCullum and Ryan Watson shared a fourth-wicket stand of 240. Watson, on 141 overnight, added just 13 but his partner carried on the battle.
McCallum's vigil at the crease lasted seven hours and three minutes, during which time he faced 254 deliveries and struck three sixes and 19 fours.
The Scots face Ireland in Belfast in five days' time in their next Intercontinental Cup clash.
