Mitch Claydon unsuccessfully appeals for lbw against Mark Chilton
Durham made Lancashire struggle with the bat on a rain-shortened first day at Old Trafford.
After the home side won the toss, they were reduced to 131-4 in the 51 overs of play that were possible.
Mitch Claydon removed openers Paul Horton and Tom Smith, while Ian Blackwell accounted for Shivnarine Chanderpaul and Steven Croft.
Mark Chilton, who was dropped on six, provided some resistance for the hosts, ending the day unbeaten on 68.
The first of three rain interruptions delayed the start by 25 minutes, but when play did begin, Smith edged Claydon to wicketkeeper Phil Mustard and Smith skewed a loose drive to point.
Chilton, who was put down at slip by Michael di Venuto off Steve Harmison, then added 42 with Chanderpaul, only for the West Indian to be trapped lbw on his home debut by Blackwell.
Croft reached 17 before he edged Blackwell to slip, leaving Gareth Cross (17 not out) to accompany Chilton to the close in an unbroken stand of 36.
Meanwhile, Lancashire second team batsman Chris Tipper, 20, is in intensive care after a car accident on Sunday evening on the way to a one-day match against Worcestershire at Ombersley.
Lancashire head coach Peter Moores told BBC Radio Lancashire: "I thought Mark Chilton played really well all the way through and Gareth Cross was nice and calm.
"It was very tough against the new ball and we saw the ball move around a little bit.
"We've still got six wickets left and the chance to build some big partnerships and hope we get ourselves in a strong position."
