Australian opener Phil Jaques was the Worcestershire matchwinner as the South Group backmarkers eased to victory at New Road for a second time in 48 hours.
James Cameron and Adam Shantry took three wickets each as Derbyshire were limited to just 147-7, of which Wes Durston was top scorer with 77.
But Jaques made short work of it, hitting an unbeaten 70 off 42 balls.
Moeen Ali weighed in with a quickfire 45 as Steve Rhodes' side won with almost five overs to spare.
While Worcestershire were winners on the field, they were losers at the turnstiles after choosing to go head-to-head with England's World Cup clash against Germany.
Only 1,000 spectators - not even half the regular turnout in this competition - were persuaded to shun the TV.
But those that did turn up enjoyed their afternoon in the Worcestershire heatwave as the home side scorched to victory.
Although still bottom of the North Division, Rhodes' men suddenly more at home in a format they have always struggled with, now Jaques and Friday night's hero Sanath Jayasuriya are firing at the top of the order.
Worcestershire now have eight days off from Twenty20 duty before heading to Old Trafford to face Lancashire next Monday night.
But they are back in Championship action at New Road on Monday when they take on Leicestershire (1100 BST).
Derbyshire are also in Championship action on Monday at home to Surrey at Chesterfield.
Worcestershire coach Steve Rhodes told BBC Hereford & Worcester: "I'm thoroughly delighted to be honest, we went through a losing run and we've always said its about momentum.
"It's a shame now that we start a four-day game tomorrow and we are just getting a grip of this and feeling confident with this type of cricket, but that's what's been put in front of us.
"I thought there were some very good performances individually but it was a good team performance as well, like on Friday night."
