Will Gidman posted career-best figures of 4-36 as Durham cruised to a 149-run win in their Clydesdale Bank 40 opener.
Gidman was backed up with the ball at the Riverside by Chris Rushworth (2-29) and Ben Harmison (2-13).
Only James Vince (27) and Hamza Riazuddin (23no) provided resistance as Hampshire were bowled out for just 115 in 23 overs.
Earlier Durham posted 264-6, Phil Mustard (74) and Harmison sharing a stand of 104 in 17 overs.
This was Durham's second victory over Hampshire in little over 24 hours, following Saturday's five-wicket victory in the County Championship.
Hampshire manager Giles While told BBC Radio Solent:
"It was a very disappointing performance. We have a lot of work to do so it will be all hands to the pump to make sure we turn things around very quickly.
"Momentum went against us early on. They started well and we never dragged it back.
"We lost early wickets and when you do that chasing a decent score it is difficult and we didn't recover."
