Stubbings and fellow left-hander Daniel Birch shared 181 for the first wicket.
In his debut in the competition Birch, who twice hit Ian Fisher over long-off for six, was caught at slip for 95.
Skipper Simon Katich hit two fours but was dismissed by Carl Greenidge, as the visitors fought back with three wickets for eight runs after tea.
Gloucestershire's attack was severely deplted, with Jon Lewis and Steve Kirby still out of action and Queensland paceman Ben Edmondson not due to arrive until next week.
Stubbings was dropped on 56 at first slip by Hamish Marshall off Mark Hardinges, but after he reached his second successive Championship century the scoring rate slowed.

edged a cut at Vikram Banerjee to slip and Stubbings fell to the slow left-armer
when he chipped to mid-on after compiling 128 from 267 balls.
With the second new ball Greenidge captured the prized wicket of Katich when the Australian was bowled offering no shot, and then Travis Birt drove at Hardinges and was caught at first slip.
