Chris Taylor's second century of the campaign could not prevent Northants taking control at Wantage Road, the home side 152 runs ahead at 92-1.
South African paceman Johan van der Wath claimed three wickets in as many overs, including New Zealand's Hamish Marshall caught at gully for a duck.
Nightwatchman Steve Kirby batted for almost three hours for 37 before Taylor shared 102 with Steve Adshead.
Monty Panesar wrapped up the innings for 250 and Northants soon ploughed on.
Glos had moments of fluency in their innings, Taylor taking two fours in an over off van der Wath and Adshead immediately sweeping Jason Brown to the mid-wicket boundary.
Although Adshead holed out to David Sales off Brown four short of his half century, Taylor reached 100 when he cut Lance Klusener for three.
Panesar had former Northants player Carl Greenidge caught at
mid-wicket by Sales, Vikram Banerjee dismissed by a brilliant diving catch by
Steven Crook at slip, and Ben Edmondson caught behind after a wild swipe.
Chris Rogers edged Edmondson's off-cutter to Adshead but Stephen Peters and Usman Afzaal put together an unbeaten 61 partnership to ensure Northants stayed in command.
