Phil Mustard helped Durham compile 58 in the first six overs but Lancashire's three spinners bowled 11 overs between them to restrict the visitors to 149-5.
Loye used his trademark slog sweep to dispatch the second ball of the innings for six but Ottis Gibson dismissed Sri Lankan star Sanath Jayasuriya cheaply.
Liam Plunkett took two wickets in his first over but Loye held firm.
Plunkett struck with his first ball to dislodge dangerous Australian Brad Hodge, then added another notable scalp with the wicket of Stuart law.
But Loye, who blasted 100 from 55 balls the last time he played against Durham in the competition in 2005 drove and pulled Plunkett for four, sharing 113 for the fourth wicket with Steven Croft, who smashed a stunning upper-cut stroke to the point fence.
