Gloucs had been dismissed for a paltry 98, Mark Hardinges top-scoring with 25 as Robin Martin-Jenkins took 3-15 and Michael Yardy 3-20.
But Sussex were stunned when Carl Greenidge removed opener Matt Prior for nine and took 4-30 with James Averis also impressing with 4-54.
The ninth-wicket pair put on 24 runs but William Rudge took 2-1.
Following his impressive spell with the ball, Martin-Jenkins revived Sussex's hopes with three fours in an over from Averis.
But he was Greenidge's fourth victim and Yasir Arafat was superbly caught at full stretch to his left by keeper Steve Snell, his fifth catch.
