The South African smashed seven sixes, the last taking the hosts past Sussex's 284-9 at the end of the 47th over, as he powered to 54 off only 20 balls.
Before that mayhem, Richard Montgomerie was the mainstay of the visitors' total, stroking 11 fours in his 127.
Martin van Jaarsveld (75 not out) and Rob Key (63) gave Kent impetus before Kemp provided a devastating flourish.
His innings made amends for dropping Montgomerie off his own bowling when the opener was on 36.
Montgomerie was run out off the last ball of the innings and things looked up for his side when Andrew Hall was beaten by Mushtaq Ahmed's leg-spin to leave Kent on 90-3.
But van Jaarsveld and Key took the game away from Sussex before Kemp's fireworks added a stunning seal to the performance which brought a fitting end to festival week at Tunbridge Wells.
He hit his maximums in the space of 11 balls - launching three out of the ground - to receive a standing ovation from the crowd.
Kemp looked forward to more one-day success after his spectacular innings condemned Sussex to their first defeat of the season.
"It was a very good pitch and I hit a few in the middle so you enjoy days like this," he told BBC Radio Kent.
"It's been a while coming, it's nice to be getting in my stride now and hopefully with Twenty20 coming up we can go from strength to strength."
