Smith, who scored a century against Surrey in the championship during the week, followed up with another to propel Kent to a huge total.
The opener's 107-ball 122 included nine boundaries and four sixes, while Mark Ealham provided some late fireworks, smashing an unbeaten 50 off 22 balls.
Mohammad Sami was the key performer with the ball as Glamorgan found an asking rate 6.5 runs per over beyond them.
Sami took three wickets, including both openers cheaply, while Robert Ferley also claimed three wickets but could not match the Pakistani for economy.
Matthew Maynard tried hard with a knock of 72, but a victory that alleviates Kent's relegation fears was never in doubt.
