Gloucester will have to settle for an away quarter-final in the European Rugby Challenge Cup after they were beaten at home by Pau, who top Pool 3.
Jeremy Thrush put Gloucester ahead but the French Top 14 side led at the break when Steffon Armitage scored from a line-out on the stroke of half-time.
Second-half tries from Frank Halai and Lourens Adriaanse eased Pau clear.
And Daniel Ramsay's score settled it in between efforts from Callum Braley and Jake Polledri for Gloucester.
Victory gave the French club six wins from six in the pool and saw them finish eight points above second-placed Gloucester, who missed the chance to take top spot.
The defeat was the Cherry and Whites' first at home in any competition so far this season and their first at home under head coach Johan Ackermann.
