Colleen Harris, 44, the Prince's deputy press secretary since April 1998, starts her new job immediately.
She was appointed after her predecessor Sandy Henney resigned in June, following a row over pictures of Prince William taken to mark his 18th birthday.
Ms Henney, who had been in the job for seven years, left after she was accused of breaking an agreement with the Daily Telegraph after releasing the photographs to other national newspapers.

"It is a great privilege and I am looking forward to getting on with the job," she said.
The Prince asked her to take over the job and no other candidates were interviewed.
Before she joined St James's Palace, Mrs Harris, who is married with two children, worked as a senior press officer in the Prime Minister's Office and the Cabinet Office.
She has also served as Head of Media Planning at the Department of Transport and the Department of the Environment, Transport and the Regions (DETR).
Two new assistant press secretaries have also been appointed - Patrick Harrison, currently assistant director in the press office at the DETR, and Kirsteen Clark, private secretary (presentation) to Scottish Secretary John Reid.
