The annual Coachella festival is set in a desert near Palm Springs and attracts a wide mix of global music stars.
Madonna will be playing in its Sahara dance tent on 30 April, while other performers include Franz Ferdinand, Scissor Sisters, Depeche Mode, the Go! Team and Yeah Yeah Yeahs.
Last month Madonna said on US TV programme the Ellen DeGeneres Show: "I cannot keep it a secret anymore. I am going on tour this summer. Are you ready?"
Madonna's last tour in 2004 saw her play 56 shows worldwide, including dates in Manchester, London and the Republic of Ireland.
Music journal Billboard said she took $125m (71m) in the process and made more money from touring than any other artist that year.
Last month the 47-year-old won the best international female artist Brit Award and had a minor operation to correct a hernia in Los Angeles.
